CellenOne X1
The CellenOne X1 is a unique platform combining high accuracy single-cell isolation and precision nanolitre reagent dispensing, allowing miniaturization and automation of single-cell proteomics protocols using off-the-shelf, commercially available consumables. As the only all-in-one instrument platform developed specifically for such assays, sample loss is kept to an absolute minimum, re-producibility across large batches kept high, and it will provide DTU Bioengineering with unprecedented analytical capabilities in the single-cell proteomics field.
DTU Bioengineering is looking to acquire the CellenOne X1 instrument, produced by Cellenion, for expanding its analytical capabilities and consolidating its internationally leading position in the field of single-cell proteomics by Mass Spectrometry (scp-MS). The instrument will be used for running large-scale scp-MS experiments, developing novel microfluidics supported sample preparation workflows, implementing single-cell multiomics experimental workflows and supporting single-cell RNA sequencing experiments.
The CellenOne X1 is the only instrument that encompasses all major aspects of scp-MS sample preparation into a single instrument, such as cell isolation, reagent dispensing and maintaining a closely controlled environment for optimal sample preparation conditions. Key features that the CellenOne provides, off-the-shelf and as an integrated instrument platform, and where it sets itself apart from all alternative products are:
1. Multi-fluorescent (blue, green, orange and red) and brightfield image-based cell sorting
2. Lossless cell sorting (collecting ‘discarded’ cells again for re-use)
3. Ultra-gentle dispensing technology
4. Cell depositing in completely controlled x,y,z plane, with axis precision <3µm
5. Dead-volume free cell depositing, i.e. down to 1µl of input sample
6. Automated single-cell proteomics preparation using off-the-shelf consumables
7. Dedicated reagent/consumable workflows for single-cell proteomics (ProteoCHIP, ProteoCHIP LFQ, Nested NanoPOTS) in nL volumes
8. Integrated temperature control, humidity control and dew-point control to prevent evaporation
9. Piezo-acoustic based picolitre to nanolitre dispensing
10. Fully customizable target plate configuration, in all 3 dimensions (x,y,z)
11. Sub-nanolitre volume cell dispensing, enabling extreme assay miniaturization and corresponding dramatic savings in the cost of reagents
12. Droplets are dispensed with microarray-type accuracy and precision with typical spot pitches of down to 100 microns in any pattern and into 96-394- or 1536-well plates or nanowell vessels or onto slides or custom substrates
13. Cells of 2 – 80 microns in diameter can be detected in IBSCITM (bright field) mode, while cells less than 1 micron can be detected using FIBSCITM (fluorescent mode)
14. Images of the droplet containing single, isolated cells and of the cells as they proceed toward the dispensed droplet portion of the nozzle are available for real-time observation and are also preserved for documentation of singularity and review of morphological characteristics.
The device utilizes, exclusively, a collection of intellectual property, including that described in the following patents:

Combined, the CellenOne is the only commercially available sample preparation platform for scp-MS, with no commercially available alternative products on the market at the time of writing.
Section IV: Procedure
The contract was awarded to Cellenion, as their CellenOne X1 product is the only instrument on the market that can fulfil all our requirements, in a single instrument platform, for 1) single-cell dispens-ing, 2) reagent dispensing, and 3) sample preparation, all in a lossless fashion. Detailed features of the CellenOne and why this is a unique instrument has been provided above.
A market survey was conducted to explore all possible options for a single instrument dedicated for single-cell proteomics by Mass Spectrometry sample preparation, and to see whether alternative options were available. We have explored different suppliers, but none of them offer a single-instrument option, with commercially available, off-the-shelf consumables for nano-litre volume lossless single-cell sample preparation while controlling sample evaporation for proteomics analysis.
Other instruments either 1) dispense single cells in too high a volume (1µL or above, whereas we need pico- or nanolitre volumes), 2) do not allow for joint fluorescent and image-based cell sorting which we require to select the correct cell types, 3) are only able to dispense either cells or reagents (not both), 4) do not provide the correct environmental control for appropriate sample preparation conditions, and 5) do not have off-the-shelf experimental workflows available for scp-MS sample preparation, meaning we would obtain insufficient data quality for our biological experiments. This makes the CellenOne X1 the only instrument currently on the market that provides a solution for our high-sensitivity and high-throughput experimental scp-MS workflow demands.

Complaint regarding prequalification:
Complaint regarding prequalification must be submitted no later than 20 calendar days after notification has been sent to the concerned Candidates informing them who has been selected, if the notification includes the grounds for the decision, cf. lov om Klagenævnet for Udbud (Complaints Board for Tenders) § 7, Section 1.
Complaint regarding award of contract:
Complaint regarding the award of a contract must be submitted no later than 45 calendar days from the date after the Contracting Authority has published a notice in the European Union Official Journal informing that the Contracting Authority has awarded/concluded a contract, cf. lov om Klagenævnet for Udbud (Complaints Board for Tenders) § 7, Section 2, no. 1.
Complaint regarding award of a contract under a framework agreement with a reopening of the competition (mini-tender):
Complaint regarding the award of a contract under a framework agreement with a reopening of the competition must be submitted no later than 30 calendar days from the date after the Contracting Authority has notified the concerned Tenderers that a contract based on a framework agreement with a reopening of the competition has been awarded/concluded, if the notification includes the grounds for the decision, cf. lov om Klagenævnet for Udbud (Complaints Board for Tenders) § 7, Section 2, no. 2.
Complaint regarding the award of a framework agreement:
Complaint regarding the award of a contract must be submitted no later than 6 months from the date after the Contracting Authority has notified the concerned Candidates and Tenderers that a framework agreement has been concluded, cf. lov om Klagenævnet for Udbud (Complaints Board for Tenders) § 7, Section 2, no. 3.
Complaint regarding direct award of contract:
Complaint that the Contracting Authority, contrary to the Public Procurement Act, has concluded a contract without prior publication of a contract notice in the European Union Official Journal must be submitted no later than 30 calendar days from the date after a contract award notice has been published by the Contracting Authority in the European Union Official Journal and that contract award notice includes the grounds for the decision to award the contract directly, cf. lov om Klagenævnet for Udbud (Complaints Board for Tenders) § 7, section 3.
Compliant regarding public procurements below the EU threshold:
Complaint regarding violation of public procurements below the EU threshold must be submitted no later than 45 calendar days from the date after the Contracting Authority has notified the Tenderers about the award of contract, if this notification includes the grounds for the decision; and no later than 6 months from the date after the Contracting Authority has notified the concerned Candidates and Tenderers that a framework agreement has been concluded, if the notification includes the grounds for the decision, cf. lov om Klagenævnet for Udbud (Complaints Board for Tenders) § 7, section 4, no. 1 and 2.
