A Crest V3 spinning disk confocal unit with Ti2-E inverted microscope frame
The Novo Nordisk Foundation Center for Protein Research (NNF CPR) is part of the Faculty of Health and Medical Sciences, University of Copenhagen. The microscope will be part of the Microscopy Platform operated by NNF CPR.
The procurement is a customized Nikon Ti2 microscope equipped with a spinning disk module coupled to a CELESTA laser engine. This configuration of the microscope is necessary for performing the technique “optical pooled screens” (Feldman et al., Nat Protoc, 2022, 17(2):476-512) . The specific combination of laser engine, custom filters, large field of view imaging, high-throughput capacity and Nis-elements/JOBS software is unique to this microscope.
Section IV: Procedure
This microscope is the only validated equipment for the technique (“optical pooled screens”) for which it will be used. The CREST v3 system has a motorized IN/OUT disk allowing a fluid widefield/confocal transitions. The system has a large field of view (25 mm diagonal) with a uniform 90% homogenous illumination over the entire 25 mm. The system comes with NIR lasers for multichannel imaging (405 / 446 / 477 / 518 / 545 / 637 / 748 nm) and a high disk speed (15,000 RPM for up to 6000 fps) which together with the slit disk optimized for high light throughput allow fast image acquisition. Finally, the system comes with software fully integrated in NIS-elements which is a powerful software suit for automated image acquisition and analysis.
This microscope is customized to enable the technique “optical pooled screens” (Feldman et al., Nat Protoc, 2022, 17(2):476-512).
