Analyzer for measuring oxygen consumption rate (OCR) and extracellular acidification rate (ECAR) of live cells in a 96-well plate format. University of Iceland, the Biomedical Center, (UoI-BMC) intend to purchase an analyzer for simultaneous real-time measurement of OCR and ECAR in living cells in a 96-well format. This will enable us for the first time to perform such measurements to obtain information on basic metabolism and mitochondrial function in real time as well as monitoring responses to test compounds and other experimentation. Importantly, the cells remain alive and can be used for further experiments and measurements. Previously we have only been able to perform point measurements on lactate and glucose concentrations.

University of Iceland, The Biomedical Center, (UoI-BMC) intend to purchase an instrument for the analysis of oxygen consumption rate (OCR) and extracellular acidification rate (ECAR) at intervals of approximately 5-8 minutes. OCR is an indicator of mitochondrial respiration, and ECAR is largely the result of glycolysis. Real-time measurements of OCR and ECAR will be made by isolating an extremely small volume (about 2 μL) of medium above a monolayer of cells within a microplate. Cellular oxygen consumption (respiration) and proton excretion (glycolysis) cause rapid, easily measurable changes to the concentrations of dissolved oxygen and free protons in this “transient microchamber” which are measured every few seconds by solid state sensor probes residing 200 microns above the cell monolayer. OCR and ECAR are calculated by appropriate software. Following baseline measurements inhibitors, stimulants, substrates, or compounds can be added by way of drug delivery ports. As the method is non-destructive, the metabolic rate of the same cell population can be measured repeatedly over time while up to 4 different drugs are injected sequentially into each well. Upon completion of the assay, other types of biological assays such as cell viability can be performed on the same plate. To maintain normal cell physiology, a temperature control system maintains the cellular environment at 37 ^oC (or other pre-set temperature).
